ตรวจหาปริมาณเม็ดเลือดขาว CD4 โดยการเจาะเลือด สำหรับผู้ที่อยู่ร่วมกับ HIV หากมีระดับ CD 4 ต่ำกว่า 200 จะทำให้มีโอกาสติดเชื้อฉวยโอกาสได้ง่าย เนื่องจากอยู่ในภาวะภูมิคุ้มกันบกพร่อง และหากผู้ที่อยู่ร่วมกับ HIV ได้รับยาต้านไวรัสต่อเนื่องจะมีค่า CD4 คงที่อยู่ที่ระดับ 500-600

ตรวจโดยการเจาะเลือด มี 2 วิธี ได้แก่
1.วิธี Anti-HIV : ตรวจหาภูมิคุ้มกันที่ตอบสนองกับเชื้อ HIV สามารถตรวจได้ตั้งแต่ 1 เดือนหลังมีความเสี่ยง
